Il 22° Congresso della Società Italiana di Tossicologia - SITOX è giunto al termine e noi di S&C BEST - Toxicology siamo entusiasti di aver partecipato a questo evento a Bologna. Durante il congresso, abbiamo esplorato il rapporto dose-effetto nella valutazione del rischio tossicologico, un tema cruciale per la nostra professione. Le presentazioni hanno coperto una vasta gamma di argomenti, permettendoci di mantenerci al passo con lo stato dell'arte, dalle nuove metodologie di valutazione del rischio, alla tossicologia ambientale e clinica. Inoltre, siamo felici di aver ampliato il nostro network e di aver stretto nuove conoscenze con colleghi e esperti del settore. Questo evento ci ha offerto l'opportunità di portare a casa nuove idee e prospettive che ci aiuteranno a migliorare continuamente nel nostro lavoro. A reminder of the EU Reg 2024/1328 about Cyclosiloxanes D4, D5 and D6. The key points of this regulation are as follows: - Restrictions on Cosmetic Products: The regulation introduces new restrictions for D4, D5, and D6 in cosmetic products. These substances cannot be placed on the market in concentrations greater than 0.1% by weight after June 6, 2026. - Environmental Concerns: The regulation aims to address environmental risks associated with the release of these substances into the aquatic environment and atmosphere. Cosmetic products, particularly leave-on products, are identified as a significant source of environmental release. - Application Date: The restrictions will start applying from June 6, 2026, allowing a transition period for industries to adapt. - Scope of Use: D4, D5, and D6 are used in various products, including cosmetics, dry cleaning, waxes, and cleaning products. The regulation extends beyond cosmetics to address broader environmental impacts. - Socio-Economic Considerations: The European Chemicals Agency's Risk Assessment Committee (RAC) and Socio-Economic Assessment Committee (SEAC) have supported these restrictions as the most appropriate measures to minimize emissions and address identified risks

Cyclosiloxanes such as D4, D5, and D6, often present as impurities (or NIAS) in silicone-based materials, cannot be evaluated using the Threshold of Toxicological Concern (TTC) approach. This method is a valuable tool for assessing the safety of chemicals with limited toxicity data. However, certain categories of substances are excluded from the applicability of TTC approach because they were not present in the initial database and therefore outside the domain of applicability. These categories include: ▶️ Organosilicon substances (as Cyclosiloxanes) ▶️ Inorganic substances ▶️ Proteins ▶️ Nanomaterials ▶️ Radioactive substances ▶️ Metals in elemental, ionic or organic form. However, in the case of organic salts, where the counter ion is an essential metal (e.g. sodium), the Scientific Committee recommends that the TTC approach could be applied to the organic ion. Other substances present special properties and for this reason are excluded from the applicability of TTC approach as well: ▶️ High potency carcinogens: aflatoxin-like, azoxy- or N-nitroso substances and benzidines ▶️ Steroids ▶️ Substances that are known or predicted to be bioaccumulative According to EFSA, 2019, those compounds cannot be evaluated using the TTC approach and a punctual toxicological evaluation might be needed. Cleaning validation is a process designed to ensure that equipment is free from any residues that could lead to cross-contamination, potentially jeopardizing the quality, safety, or efficacy of products. A crucial aspect of cleaning validation involves setting safe limits for these residues, known as Permitted Daily Exposure (PDE). This term refers to the maximum amount of a substance that an individual can be daily exposed to, without experiencing adverse health effects. The PDE is specific to each substance and is determined by a toxicologist using international guidelines and scientific methods. Once established, the PDE is utilized by cleaning validation specialists to determine the Maximum Allowable Carry Over (MACO) value. This indicates the acceptable level of residual product that can be transferred from one batch to another without compromising the safety and quality of the subsequent product. For pharmaceutical manufacturing facilities, Contract Manufacturing Organizations, and the food industries, deriving and establishing the PDE is a vital component of cleaning validation programs to meet various regulatory and current Good Manufacturing Practice (cGMP) requirements.
