Kicking off the new year with some really exciting news – our review on #Nature Review Drug Discovery “Targeting mitophagy in neurodegenerative diseases" is finally out! It’s been an amazing journey working on this. Huge thanks to Laura Parton, Paul Thompson, Nicholas Hertz, Ph.D and Miratul Muqit for the collaboration, their incredible insights, and for making this a truly enjoyable experience. 🧠🧵Mitochondrial dysfunction is a hallmark of many idiopathic neurodegenerative diseases, including Parkinson's, ALS, Alzheimer's, and Huntington's. In familial forms of Parkinson's and ALS, mutations in genes linked to mitophagy deficits highlight a key pathway that could hold therapeutic potential. In our review, we explore the evidence supporting enhancing mitophagy—the process by which damaged mitochondria are cleared from cells—as a novel therapeutic strategy. This approach offers the potential to deliver neuroprotection and disease modification, addressing a critical unmet need in the treatment of neurodegenerative diseases. Great progress is being made with small-molecule approaches targeting mitophagy, like USP30 inhibitors and PINK1 activators, now entering phase I clinical trials!
