Refeyn, the pioneer of mass photometry, is delivering a disruptive new generation of analytical instruments that let you weigh individual molecules with light. Mass photometry offers new possibilities to characterise the function, structural composition and dynamics of biomolecules. Refeyn instruments measure the mass of individual molecules directly in solution, rapidly, and simply, revealing the true behaviour of molecules in near-native environments, providing scientists with key information about their mass distribution. Refeyn was spun out of Oxford University in 2018 to make mass photometry available globally. Its technology has now been rapidly adopted across academia and industry, where it is transforming biomolecular characterisation.

    Arvind Pillai’s talk on protein design is almost here!🔬 A researcher in the lab of 2024 Nobel Laureate David Baker, at the Institute for Protein Design, University of Washington, Arvind will present his work on the design of proteins de novo using computational tools💻🧬.   • When it comes to characterizing protein oligomerization, #MassPhotometry really excels🔬, and his has been shown again in a recent paper – this one explores the structure and biochemistry of a fungal Group 5 septin.  Septins play important roles in membrane organization, polarization and morphogenesis🧬. Group 1-4 septins are well studied, as they are present in humans and yeast, but other groups are not.  Grace Hamilton, Katherine Wadkovsky and Amy Gladfelter – from the University of North Carolina at Chapel Hill and Duke University – characterized the KpAspE Group 5 septin from Knufia petricola, finding that it recapitulates many of the properties of more well-known Septins.  Using mass photometry, they showed that KpAspE formed mainly monomers and dimers at nanomolar concentrations, but its oligomerization behavior was pH-dependent, with trimers appearing at pH 6.4 and dimers becoming more dominant at pH 8.4.  Read the full paper here! 👉 https://lnkd.in/dWzGGE39 #MassPhotometryStories

  • Peter Eisenhut and colleagues from Takeda analyzed rAAV9 samples produced with two different transfection reagents: FectoVIR and Polyethylenimine. Biopotency assays found that rAAV9 samples produced with FectoVIR had higher potency. Mass-based assays like mass photometry and AUC measured the proportions of empty, full and partially filled capsids and were in excellent agreement (😉), but did not detect differences between transfection methods. On the other hand, sequencing showed that the proportion of full-length transgene loads was lower than expected from mass-based methods, and did find a difference between transfection reagents, in agreement with biopotency assays.
