APP NOTE: Proteins play a crucial role in regulating cellular activities, including metabolism, signal transduction, structural support, and gene expression. Understanding protein-protein interactions (PPIs) is essential for drug discovery, as it can reveal novel targets for modulating disease pathways. However, less than 5% of potential PPIs have been characterized. In this app note, we describe how #HuProt™ Microarray addresses this gap by featuring over 21,000 complete human proteins and protein isoforms, covering more than 81% of the human proteome. This unique technology allows researchers to analyze PPIs across the entire human proteome in a single experiment, facilitating a comprehensive understanding of interactions related to specific proteins. APP NOTE: #HuProt™ Proteome Microarrays are fabricated by immobilizing 21,000+ individually purified GST-fused human proteins on a single glass slide. Unlike nucleic acids, many proteins can become unstable after purification; therefore, correct folding of proteins prior to immobilization on a solid surface is crucially important for maintaining their physiological activities. The choice of protein expression system greatly influences the folding and post-translational modifications of proteins and can affect the success rate of protein purification. In this app note, we explain the advantages of a yeast expression system when compared to E. coli and cell-free expression systems. In short, a yeast system provides a eukaryotic environment conducive to proper protein folding and stability, with expression of necessary chaperones and co-factors to prevent aggregation and support translation of large molecular weight proteins. APP NOTE: The #VirD™ Membrane Protein Microarray represents a groundbreaking platform for screening biologics against membrane proteins. The VirD technology consists of HSV-1 virions which are engineered to display membrane-embedded proteins of interest with native folding and function. Each engineered virion is individually cultured in human cells, purified, and printed onto glass slides to create VirD microarrays. In the app note, we describe how embedding functional, post-translationally modified receptors in the lipid envelope of viral particles allows VirD to preserve the native conformation of GPCRs, enhancing receptor functionality and data accuracy. This method also eliminates interference from endogenous proteins, often seen in cell-based systems, and provides unbiased screening for targets, even when the downstream signaling pathways are unknown.
