A new mechanism of regulation of host responses to SARS-CoV-2 infection has been uncovered. Our study in Cell Reports by Cell Press shows that the SARS-CoV-2 main protease 3CLpro is unconventionally secreted from infected cells via gasdermin pores. Extracellularly, the protease reduces platelet activation and inactivates antiviral interferon-lambda 1. This finding reveals a novel phenomenon of virus protease secretion and new functions of a viral enzyme outside of host cells. This work was performed for my PhD thesis in Christopher Overall's lab at The University of British Columbia, including key collaborations with Arinjay Banerjee and Kaushal Baid at Vaccine and Infectious Disease Organization (VIDO) and University of Saskatchewan; Charaf Benarafa, Nedim Kozarac and Nathan Leborgne at the Institute of Virology and Immunology IVI and University of Bern; Hugh Kim and Steven Zhexuan Jiang also at UBC; and John Vederas at University of Alberta.
