Expansion #Microscopy #ExM is a versatile #SuperResolution technique for fixed samples, but the complete ExM sample preparation process can require days to perform. New research shows how ExM sample prep can be performed ~15X faster using microwave radiation to accelerate/enhance both the fixation and immunofluorescence staining processes. The authors demonstrate their approach in both X. laevis and D. melanogaster brain tissue. Click to view the #OpenAccess article: https://bit.ly/4izlJvj #Microscope #Imaging #SciComm

Fluorescent biosensors are pivotal tools for quantifying intracellular dynamics. While readout is often based on the intensity of emission, other properties, such as fluorescence lifetime, offer different advantages. New research introduces a family of four biosensors (for ATP, cAMP, citrate, and glucose) based on the #FluorescentProtein mTurquoise2 and using a common design framework. These biosensors are dual functional – compatible with readout using both intensity-based imaging and lifetime-based imaging with fluorescence lifetime imaging #Microscopy #FLIM. Renal glomeruli help make up the basic filtration unit of the kidney, however their small size and complex structure can make their structure difficult to interrogate using diffraction-limited optical imaging. A new preprint article introduces a pipeline using #SuperResolution expansion #Microscopy #ExM for 3D imaging of whole glomeruli and #MachineLearning-based annotation and analysis of glomerular features. #Imaging of expanded samples for this study was performed using a Nikon A1R #Confocal system with 40X/1.15 LWD water immersion objective.
