Influence of Degrader−Antibody conjugates (DACs) The targeted protein degradation (TPD) technology employing proteolysis-targeting chimeras (PROTACs) has been widely used to treat cancer and other diseases. Targeting undruggable targets and overcoming drug resistance have been demonstrated significant advantages of PROTACs. Due to their substantial molecular weight, PROTACs still face challenges related to membrane permeability, despite releasing targeted proteins efficiently. Furthermore, off-target toxicity must also be addressed. In order to address these issues, Degrader-Antibody Conjugates (DACs) have been developed, which take advantage of the targeting and internalization capabilities of antibodies.

Excited to share a recent publication by my mentee and colleague, Dr. Nishika Karbhari at The Mayo Clinic describing the use of marine-derived therapeutics for #glioblastoma! Here are some of the highlights: - Sea cucumbers, seagrass, sea urchins, and sea squirts all have bioactive compounds that have anticancer properties - Compelling activity has been shown to modulate the tumor immune microenvironment - Various antibody-drug conjugates rely on marine-derived chemotherapeutic backbones for activity - Holothurian triterpene glycosides (sea cucumbers) and fucoidans (seagrass) warrant further investigation as potential lead compounds

#ESMOVirtualPlenary: Findings from a first-in-human phase I study investigating IBI363, a PD-1/IL-2 bispecific antibody fusion protein, provide proof-of-concept on its capacity to revert acquired resistance to immunotherapy in pre-treated patients and in some populations with a poor prognosis including patients with #MucosalMelanoma. According to Alberto Hernando Calvo additional work remains to be conducted to fully elucidate the subset of patients who may derive benefit from IBI363, but indeed the data presented support further investigation to pave the way for designing novel IL-2-based immunotherapy combinations in the anti-PD-1/L1 resistant setting.
