The recent study by He et al. (2024) explores the therapeutic potential of UC-MSCs and their extracellular vesicles (EVs) for treating chemotherapy-induced premature ovarian failure (POF). Administering UC-MSCs or EVs on day 6 after chemotherapy significantly improves ovarian function by promoting follicle growth, enhancing granulosa cell proliferation, and inhibiting apoptosis. EVs derived from UC-MSCs activate the PI3K/AKT pathway, with clusterin (CLU) playing a key role in reducing apoptosis. This study highlights a new molecular mechanism for UC-MSCs in POF treatment, offering a promising cell-free therapeutic approach to restore ovarian function. An interesting study coming out of Thailand. Cannabidiol suppresses proliferation and induces cell death, autophagy and senescence in human cholangiocarcinoma cells via the PI3K/AKT/mTOR pathway while this is a bit thicker in context. Recently, I participated in clinical research training for a phase III study of givinostat in polycythemia vera (PV). Givinostat represents an innovative approach to PV treatment, differing from standard therapies like hydroxycarbamide. It targets cell proliferation by inhibiting cells with the JAK2 V617F mutation, prevalent in PV patients. Early phase trials have shown givinostat's effectiveness, even in patients unresponsive to hydroxycarbamide, highlighting its potential as an alternative or supplementary treatment for PV. 🥼 🔬 🧪 Paper of the Day ❗ ❗ ❗ Activating the NFE2L1-ubiquitin-proteasome system by DDI2 protects from ferroptosis A Paper of the Day deals with a study published in Cell Death & Differentiation sheds light on the complex interplay between ferroptosis, a type of cell death, and the NFE2L1-ubiquitin-proteasome system. The researchers found that ferroptosis can lead to the activation of NFE2L1, a transcription factor involved in regulating the proteasome, a cellular machinery responsible for protein degradation. This feedback loop helps cells cope with oxidative stress and prevent further damage. The study also identifies DD12, an aspartyl protease, as a key regulator of this process. DD12 is responsible for cleaving NFE2L1, which is essential for its activation.
