Last week, I attended the #spatialomics24 conference in the beautiful city of Ghent. While I was highly impressed by the intellectual effort and hard work invested in developing new technologies, analysis pipelines, and AI tools, I started to wonder about the field’s ultimate contribution to discovering new biology. Undoubtedly, there were some novel findings, but they often came at a high cost and could, in the end, be recapitulated by simple antibody staining. For example, different zones in tumors discovered by multi-omics techniques were already visible on the H&E slide. High-dimensional omics datasets were reduced back to basic cell types (what are hundreds of transcripts worth if at the end I just use CD8 expression?). AI was used to generate images from UMAPs (but to what end?). Don't get me wrong, the field is still in its early stages, and promising technologies are emerging daily. I was particularly impressed by label-free tools, including Raman imaging. However, we might need to refocus on the biology itself more frequently. We should start testing hypotheses in real experiments and re-examining what constitutes correlation versus causality. And this might mean to include reporter and recording systems into the OMICs pipeline such as LIPSTIC (labelling immune partnerships by SorTagging intercellular contacts), fate-mapping/barcoding with Polylox/Polytope or timers such as Zman-seq.

As 2024 winds up, I wanted to share @michael levinTufts University  and his work on bioelectricity, OpenFold, an open source AI biology consortium and my excitement on quantum computing.   Let’s kick it off with Michael Levin. I came across him and his work on bioelectricity and mammalian regeneration through the book “We are Electric”. His research on how bioelectricity provides the blueprint for cells to communicate could have profound impact on regenerative medicine. If this piques your interest, you can find out more here Morphoceuticals   It wouldn’t be 2024 without a mention of AI. But I would like to spread awareness of OpenFold an open-source consortium democratizing AI for biology and drug discovery with protein structure prediction training and inference models.   While on the topic of AI, Quantum Computing (QC) deserves a mention. One of the challenges in human health is our understanding of human biology. To get a sense of the issue, to date less than 5 (sourcehttps://lnkd.in/dsxrxYvT) of hundreds of thousands of human protein interactions have been mapped. Through superposition and entanglement, QC provides the computational scale that can save years of iteration and improve the success rate of drug discovery.   There are other research areas such as a game changing breast cancer detection device, an innovative approach to drug delivery and engineered macrophages respectively that are too early to highlight but I am looking forward to the strides they make in 2025.
