Everyone has heard about Penicillin. And many would remember how a sharp-eyed Alexander Fleming discovered it. Perhaps a few would even remember that penicillin was extensively tested as a drug by Howard Florey and Earnest Chain along with Norman Heatley who helped produce sufficient material for the testing. But did you know that their first test subject actually died? Not because the drug wasn’t effective but because they ran out of it! And this is where Chemical Engineers led the effort to provide the world with its first commercial antiobiotic that too, during WW2. The Challenge of Scale-up Initially, everyone favored chemical synthesis over fermentation. This was for a variety of reasons 1️⃣ Chemical synthesis was an already established process and could be made continuous, allowing for greater production while also being faster. 2️⃣ Fermentation has a low rate kinetics and therefore, a larger reactor vessel would be needed for similar production. 3️⃣ The concentration of penicillin in the final broth was very low and therefore, purification became complex. And penicillin was a relatively unstable compound limiting the choice for separation and purification. The solutions were just as important as penicillin itself ▶ To improve yield, labs worldwide searched for a better producer strain for greater yield of Penicillin ▶ Submerged Fermentation: A younger method of fermentation where the producer strain was allowed to grow in a liquid broth medium. The previous method of growing strain on a solid substrate proved less effective and was eventually discarded. In the end, Pfizer developed production capacity to cover the penicillin demand during the WW2. The Key Takeaway The scale-up of Penicillin production was a joint multidisciplinary effort and is a great testament to the importance of chemical engineers whenever there is a need for ramping up production.
