Organoid models have accelerated the development of drug screening methodologies. However, it is important to be wary of phototoxicity. Dr Valeria Chichagova, PhD, Director of Technology, spoke to Biocompare to share some top tips on minimizing or eliminating phototoxicity: 🔹Utilize low-intensity illumination by using the lowest possible light intensity that still provides clear images to reduce cellular stress and damage. 🔹By optimizing imaging protocols to limit the duration and frequency of light exposure, researchers can further reduce exposure time. Furthermore, near-infrared fluorophores are less damaging to cells compared to visible light fluorophores. 🔹Advanced imaging techniques, such as light sheet fluorescence microscopy offers gentle, low-phototoxicity imaging, ideal for long-term studies. 💬 “Advanced algorithms for image analysis enhance the ability to extract quantitative data from complex organoid structures, improving the accuracy of phenotypic assessments. Ex vivo, before clinical study can perform out of the living, the procedures performed on tissues or organs extracted from a living organism and conducted outside the original biological context. These experiments are conducted in a laboratory setting, where the biological material is maintained in conditions that mimic those found within the organism as closely as possible. A simpler ex vivo definition would be taking a piece of a puzzle to look at it closely. Ex vivo models are frequently used for disease modeling, Drug Discovery and Development, regenerative medicine, and tissue engineering. Ex vivo and in vitro models represent an excellent alternative to animal testing.
