I really enjoyed sharing our work on Pre-Targeting with in vivo click chemistry. Many recognize that off-target toxicities are dose-limiting the vast majority of our ADCs and the oportunity to dose past this will likely lead to more benefit for patients. We know the problem is that <1% of the ADC dose goes to the tumor and the rest is catabolically eliminated in normal tissues leading to off-target toxicities. How can we really call these targeted therapies? We need to actually target them with a pre-targeting approach and reduce these off-target toxicities. Not solving this is holding back so much technology in the field. This needs to be the primary focus of the field!
