Our new chapter on #ketamine and #esketamine is out! It's hard to believe that 7 years have passed since Sanjay and I published the previous version in the APA Textbook of Psychopharmacology, 5th edition. So much has changed in that time and, while we've done our best to capture how this clinical literature has evolved, there are also certainly areas we will have missed. This is ultimately a testament to the amount of inquiry and innovation happening in the ketamine, #dissociative therapy, and related #psychedelic therapy space. Here we review information on the history and discovery of ketamine, preclinical and clinical pharmacology, pharmacokinetic parameters, mechanism of action, indications and efficacy, side effects and toxicology, and drug-drug interactions. Other chapters follow the same format, each examining the drug classes that form the basis of modern psychopharmacological treatment. Wow!! Congrats David and Sanjay. You two are the brightest stars in the galaxy of psychiatric thought leaders. Well done! Ketamine and esketamine (Spravato) are both hugely innovative and often life saving medications. A full chapter such as this in the major text book on psychopharmacology is a very positive development.

Understanding their pharmacology is critical for advancing drug development and medical research. In this application note, we explore how time-resolved impedance measurements offer a non-invasive, label-free approach to studying GPCR activation and signaling in real-time. This innovative method provides continuous cellular monitoring, delivering valuable insights into GPCR pathways. The app note highlights findings from two key pathways: 🔹 𝗛𝗶𝘀𝘁𝗮𝗺𝗶𝗻𝗲 𝗛𝟭 𝗥𝗲𝗰𝗲𝗽𝘁𝗼𝗿 (𝗛𝟭𝗥): Gq-protein coupled, involved in phospholipase C activation. 🔹 𝗡𝗲𝘂𝗿𝗼𝗽𝗲𝗽𝘁𝗶𝗱𝗲-𝗬 𝗥𝗲𝗰𝗲𝗽𝘁𝗼𝗿 𝟰 (𝗬𝟰𝗥): Gi-protein coupled, influencing adenylate cyclase and cAMP levels. The RCPA has developed new resources in partnership with the Australasian Society of Clinical and Experimental Pharmacologists and Toxicologists (ASCEPT) and the Pharmaceutical Society of Australia (PSA) to provide consensus-based guidance for Australian prescribers, pharmacologists and pathologists about drugs for which pharmacogenomic testing may be indicated. This project, funded by Australian Genomics, provides clinical indications for 35 medications for pharmacogenomic testing in Australia with the aim of improving medicine use by increasing efficacy and avoiding adverse reactions.  The project formed a multidisciplinary advisory group with experts in pathology, pharmacology, pharmacogenomics and prescribing practice from the RCPA, ASCEPT, PSA, University of Sydney, University of NSW, Monash University, University of Adelaide, University of Queensland and general practice. #pathology #pharmacology #pharmacogenomics https://lnkd.in/g-mYh_x4
