#CustomerPublicationWeekly Erectile dysfunction (#ED) significantly impacts male #fertility and quality of life. A recent PNAS paper (https://lnkd.in/d84iZmgg) describes an innovative magnetic soft microrobot comprising an ultrasoft hydrogel microsphere embedded with a magnetic nanoparticle chain for mesenchymal stromal cells (#MSCs) delivery which shows significantly enhanced MSC retention and survival. In both rat and beagle ED models, MSC-Rob treatment accelerated the repair of corpus cavernosum tissue and restored erectile function. #Single-cell RNA sequencing (scRNA-seq) revealed that MSC-Rob treatment facilitates nerve and blood vessel regeneration in the corpus cavernosum by increasing the presence of regenerative macrophages. Singleron's GEXSCOPE Products were used in this study.

In vitro maturation (IVM) is one of the commonly used artificial reproductive techniques in clinics, originally used to simulate the in vitro maturation process of oocytes. In this study, Wei Wu and team in Nanjing, use mice IVM to explore the effect of putrescine on the in vitro maturation of oocytes from reproductively old mice. Key information findings in the paper show that Putrescine: • Promotes nuclear homeostasis and cytoplasmic maturation of oocytes from reproductively old mice • Can reduce DNA damage and improve mitochondrial function of oocytes from reproductively old mice • Improves oxidative stress of oocytes from reproductively old mice by promoting autophagy • Promotes the localization of p53 in the nucleus, activates AMPK transcription and regulates oxidative stress of oocytes from reproductively old mice. 💊🔬 Current treatment option for #ChestWallEwingSarcoma. 🔬💊 Chest wall Ewing sarcomas, historically known as skin tumors, are tumors formed by a particular type of bone or soft tissue cell. It is often found in children, adolescents, and young adults with a female preponderance. The cause of Ewing sarcoma is not known. Most commonly, they develop on the ribs, but they can also occur on the paravertebral, sternal, and scapular areas. Chest wall Ewing sarcoma is a very rare and one of the most aggressive malignancies, with 5-year survival rates ranging between 40 and 60%. This week's #onlywithHCR spotlight highlights the Kozareva et al. 2021 publication, "A transcriptomic atlas of mouse cerebellar cortex comprehensively defines cell types." Kozareva et al. implemented #HCRimaging to compare neuronal population analyses in the cerebral cortex in both mouse and human. This analysis found that "...considerably more regional specialization in PCs—especially in posterior lobules—than was previously recognized. These PC subtypes overlap with greater local abundances in UBCs and in distinct specializations in granule cells, which indicates a higher degree of regional circuit heterogeneity than previously thought." #Nature: Breakthrough in #Ferroptosis Reveals Its Intercellular Propagation Mechanism! Previous studies have shown that ferroptosis can spread from one cell to another, leading to large-scale #celldeath. However, the mechanism and action of this propagation process have been unclear. On July 10, 2024, a team from China published in #Nature: Emergence of Large-Scale Cell Death Through Ferroptotic Trigger Waves. They developed a new method to isolate and study the intercellular propagation of ferroptosis, providing new insights into large-scale cell death and revealing the significant role of ferroptosis in embryonic development. The study demonstrates that #ferroptosis propagates through ROS-triggered waves, enabling large-scale cell death within cell populations. It also uncovers potential mechanisms of #ferroptosis in cellular remodeling and organ formation, offering new perspectives for exploring the interaction between developmental signals and #ferroptosis. The weeks #fridayread gives us some valuable insight into the relative mystery of the development of the mammalian body plan during gastrulation. Sato and Rosa et al. have developed and characterized a 3D culture of self-renewing mouse embryonic cells that captures the main transcriptional and architectural features of the early gastrulating mouse epiblast. In the lab of Marta Shahbazi, a group leader at MRC Laboratory of Molecular Biology (LMB) , they found that proliferation-induced crowding triggers delamination of cells that expressing high levels of atypical protein kinase C (aPKC). aPKC has previously been shown to play a critical role in cell polarity. They found that cells then became more sensitive to Wnt signaling and upregulated the expression of primitive streak markers such as Brachyury. This co-ordination of ingression and differentiation ensures that the right cell types become specified at the right place during embryonic development.
