Thrilled to share my #RSCPoster for online RSC Poster Conference 2024 on LinkedIn platform entitled as "Ionic-molecular organocatalysis enabling allylic amination reactions". We report here an "ionic-molecular" organocatalysis as a viable substitute for the metal-dependent allylic amination reactions. Mechanistic highlights supported by 31P nuclear magnetic resonance, high-resolution mass spectrometry, and experimental studies highlight the prior covalent ligation of allylic alcohols via ionic catalysis coupled with nucleophilic phosphine catalysis, yielding allylamines via the SN2’ -SN2’ (or SN1-SN2’) pathway. Implementation of this ionic-molecular catalysis in heterocyclic functionalizations (N-allylation) with excellent chemo- (N vs. O), regio- (linear vs. branch), and stereo-selectivity (E vs. Z) and active pharmaceutical ingredient (flunarizine, cinnarizine, and naftifine) synthesis has been successfully demonstrated. #RSCPoster #RSCCat #RSCOrg

A new paper released, titled "Is the Functional Response of a Receptor Determined by the Thermodynamics of Ligand Binding?" explores the Absolute-Binding Free Energy Perturbation (ABFEP) method's utility in predicting ligand efficacy across a broad spectrum of target-ligand pairs, chiefly focusing on G protein-coupled receptors (GPCRs) and a nuclear receptor. The methodology hinges on selecting ligands with available functional data, encompassing both congeneric series and diverse molecular frameworks. Researchers employed Molecular Dynamics (MD) simulations to generate template conformational ensembles reflecting both the active and inactive states of receptors. Notable elements of their approach include k-means clustering of Cα atom distances within the binding pocket, the creation of initial structures, and the application of restraints for Free Energy Perturbation (FEP) calculations. They employed a flat-bottom harmonic potential to restrain the receptor's Cα-atoms to refine their predictions. The results showed an impressive initial prediction accuracy of 93% across the examined target-ligand pairs using ABFEP, which was further enhanced to 98% by fine-tuning the classification thresholds for each receptor. This high predictive success is attributed to the method's ability to calculate the differential binding free energies between ligand interactions with the active and inactive receptor states. The research underscores that a receptor's functional response is dictated by the equilibrium between its conformational states rather than the binding affinity to a single state. This study highlights the critical role of accurately defining restraints and ligand positioning within the receptor's binding sites to achieve reliable predictions. https://lnkd.in/gFs7TkMz
