Understanding how secretory cargo is transported through the Golgi stack is one of the most fundamental and pressing questions in the field of membrane trafficking. During my postgraduate years, the membrane trafficking community was deeply divided between the maturation and stable compartment models. This intense debate left a lasting impression on me, motivating me to pursue research on the Golgi when I established my own lab. Over the past ten years, we have developed several innovative tools that allow us to directly and quantitatively visualize intra-Golgi transport of secretory cargos. Our first major endeavor in this direction has been published in eLife, where we directly measured the intra-Golgi transport kinetics of 11 different secretory cargos. Our data suggest that the Golgi operates with a stable compartment nature, supporting a modified version of the stable compartment model known as the rim-progression model. https://lnkd.in/gYm6f3MX
