#Researchers, guanosine triphosphate cyclohydrolase 1 (GCH1) activity is allosterically regulated by phenylalanine and BH4, but with opposite effects. Elevated phenylalanine levels lead to stimulation of GCH1 activity, whereas BH4, the end product of the biosynthesis pathway, inhibits GCH1 via a negative feedback loop. Comprehensive structural studies led to the discovery of AXSP0056, an allosteric molecule that selectively occupies the BH4 binding site on GCH1, thereby inhibiting its activity. In this poster presentation, we discuss how we developed and validated a sensitive LC-MS/MS assay for atropine, quantifying it across a wide range (80 pg/mL to 40 ng/mL) in rabbit plasma. Our method addresses the challenges of enantiospecific and species-specific atropinesterase, ensuring accurate achiral quantitation. Following ICH M10 guidelines, we achieved a 100% success rate in incurred sample reanalysis, confirming the robustness of our sample stabilization strategy. Congratulations to Yaowen Wu (Umeå Univ., Sweden), Herbert Waldmann (Max-Planck-Institute, Dortmund) and their research teams, for the work by A.Knyazeva et. al just published in PNAS "A chemical inhibitor of IST1-CHMP1B interaction impairs endosomal recycling and induces noncanonical LC3 lipidation"! Proud that our Chemical Proteomics Unit at the Karolinska Institute has supported such a journey with our MS-based proteomics target ID and MoA elucidation methods, particularly, with our PISA method to identify IST1 as the target of Tantalosin, and with HDX (H/D exchange) MS to map the Tantalosin binding site and the allosteric site impairing IST1-CHMP1B interaction upon binding.
