#Researchers, guanosine triphosphate cyclohydrolase 1 (GCH1) activity is allosterically regulated by phenylalanine and BH4, but with opposite effects. Elevated phenylalanine levels lead to stimulation of GCH1 activity, whereas BH4, the end product of the biosynthesis pathway, inhibits GCH1 via a negative feedback loop. Comprehensive structural studies led to the discovery of AXSP0056, an allosteric molecule that selectively occupies the BH4 binding site on GCH1, thereby inhibiting its activity.
