Our spheroid-on-chip technology in action This video contains a summary, of our project with Prof. Dr. Nicole Teusch and Alina Deipenbrock from the University of Düsseldorf, about the development of an immune competent PDAC biochip model for preclinical testing. Within this project, we have a developed an innovative biochip with three culture chambers. This biochip enables the cultivation of complex, three-dimensional tissue cultures and opens up the possibility of cultivating spheroids and stem cell-based organoids in one chip platform and to combine them with other tissue components, such as blood vessel tissue. In this video they explain their research on identifying fungi which are a rich source of potential active substances and what methods are being used to test the identified substances in their laboratory. Novel therapies are needed for Pancreatic ductal adenocarcinoma (PDAC). #PDAC is among the deadliest solid malignancies, with a five-year survival rate of less than 10%. Classically, drug screenings were performed on cultured pancreatic #tumor cells. However, such 2D-models showed limited translation relevance as they fail to recapitulate the cellular heterogeneity, structural complexity, and vascularization of PDAC tumors. We are developing a three-channel micro cavity-equipped microfluidic #biochip to generate patient-derived pancreatic cancer spheroid-on-a-chip models for screening novel therapeutic approaches. Here is a summary of the current outcome of this project: / The Dynamic42 3-chamber biochip allows controlled spatially separated coculture of spheroids and endothelial cells / PDAC spheroids in combination with perfusion strongly impact endothelial cell fitness / Integration of a CAF barrier promotes survival of the endothelial cell layer / We are now using this system to screen for novel PDAC therapeutics that are administered through the vasculature

New research alliance Together with Universitätsklinikum Jena we are working on the development of a stem cell-based liver model (Vascu Chip). The result will be a biochip-based model for personalized drug testing. The network is funded by the Thüringer Aufbaubank with #EU and #EFRE funds (FTI, Project nb. 2024 VFE 0050). Our scientist, Joanna Nowacka will be working on the vascularized liver-on-chip model for personalized compound testing. Within her work, Joanna will be developing stem cell cultures, from which she can then differentiate multiple cell types. Joanna says: “I believe organ-on-chip technology represents the future of drug testing models. I am eager to join Dynamic42 to contribute my stem cell expertise to their already remarkable platforms.”
