Once thought to be a cannabinoid receptor, GPR55 has emerged as a distinct target with unique ligand recognition and signaling mechanisms. This study identifies lysophosphatidylinositol (LPI) as the authentic ligand for GPR55 and demonstrates its signaling through the G13 pathway ( https://loom.ly/PbCWLUE ). Using cryo-EM, researchers resolved high-resolution structures of LPI- and AM251-bound GPR55, revealing a binding mode distinct from cannabinoid receptors.
