Ahead of print Acute kidney injury during transplantation and the role of inflammasomes: A brief review The increasing demand for an effective therapeutic modality in the form of organ transplantation leads to the need to improve the long-term outcomes of the process. Ischemia/reperfusion injuries (IRI) are an integral part of the kidney transplantation process, contributing to inflammation, oxidative stress and activation of the immune system. Inflammasomes, as a component of the immune response, in the form of inflammatory mediators during infection or tissue damage, initiate cell death called pyroptosis. In this context, we have defined the process of inflammasome activation in response to IRI, which is a potential cause of early kidney rejection due to increased susceptibility of the kidneys to ischemia. This review focuses on analyzing the modulation of inflammasome activity in kidney transplants and the activation of the NLRP3 inflammasome as a crucial element of kidney injury during the transplantation procedure, which could be a potential target for future preventive/therapeutic strategies. https://lnkd.in/d8knd2Qv

📰 We are thrilled to share our latest publication: "New insights in immunomodulation for intestinal transplantation." 📌 Achieving tolerance in solid organ transplantation remains a significant challenge. This is especially true for intestinal grafts due to the small bowel's high immunogenicity. This review highlights the foundational chimerism theory introduced by Thomas Starzl, provides an overview of our current understanding of tolerance in intestinal transplantation, and outlines recent advances in the field. Title: Continued hypothermic and normothermic machine perfusion without a perfusion solution switch S.B. Bodewes Summary Liver transplantation is the only treatment for end-stage liver disease, but donor shortages necessitate expanding donor criteria. Sequential hypothermic and normothermic machine perfusion make it possible to asses the viability of high-risk donor livers. However, this protocol suffers from additional cold ischemia time and complexity, limiting its widespread adoption. To address these issues, we propose investigating a modified protocol aiming to streamline the process and increase accessibility for transplantation centers globally. Our research objectives are to demonstrate the safety and feasibility of an alternative DHOPE approach that eliminates the need for the perfusion fluid exchange. The exchange is deemed necessary because the red blood cells used during warm perfusion can not be used during hypothermic machine perfusion. Expected results include outcomes in safety and feasibility through pre-clinical studies on pig livers and validation with human research livers.

We all know anemia is a common disorder, but have you heard of Fanconi Anemia? Unlike typical anemia, Fanconi Anemia is a rare genetic condition that cannot be managed by medication. The only treatment option available is bone marrow transplant (BMT). However, the success of BMT depends on proper pre-transplant planning and post-transplant care.
