I'm thrilled to share our latest bioconjugation paper, helmed by Andres Rocha Tapia and Fabrice Abgottspon, now published in Chemical Science (one of my favourite journals!). Andres and Fabrice were fantastic throughout this project and I could not have asked anymore of them. I feel very privileged to have worked with them on this and know the work would not have been possible without them. This work demonstrates the superiority of site-directed conjugation approaches vs traditional non-selective chemistries for constructing affinity protein–ssDNA conjugates. We show that conjugates constructed from the site-selective chemistries display improved specific target binding and decreased non-specific binding, including for cell membrane targets. We hope this work can convince others to take a more considered approach when constructing affinity protein–ssDNA conjugates, particularly for analytical applications such as immunoPCR and proximity ligation/extension assays (PLA/PEA). I want to take this opportunity to thank Per-Åke Nygren and Johan Nilvebrant for their help with the SPR, and Vijay Chudasama for help on the bioconjugation. Available for #License: Emory University researchers have created a collagen substitute using synthesized collagen-mimetic peptides (CMPs) to overcome contamination and immunogenicity issues in traditional animal-sourced #collagen. The #peptides form stable triple helices and, under specific conditions, assemble into nanotubes with defined inner and outer surfaces based on their peptide sequences. Conventional solid-based SERS substrates often face challenges with inconsistent sample distribution, while liquid-based SERS substrates are prone to aggregation and precipitation, resulting in irreproducible signals in both cases. In a collaborative study with Dr. Zhang in Lanzhou University, we tackled this dilemma by designing and synthesising raspberry-like plasmonic nanoaggregates that exhibit a high density of hotspots and are colloidally stable at the same time, allowing ultrasensitive detection of pollutants and biomarkers at trace concentrations.
