Calprotectin levels are measured to diagnose and differentiate between inflammatory bowel diseases (i.e.-Crohn’s disease / ulcerative colitis) from non-inflammatory conditions (i.e.-irritable bowel syndrome). They have an extensive range of applications in microbial biotechnology and diagnosis processes. The abnormality of the enzyme metabolism system leads to many metabolic diseases. Further, enzymes can be used as medicines to treat a range of diseases; they are called medicinal enzymes. For example, trypsin can be used not only to accelerate wound healing and dissolve blood clots but also to remove necrotic tissue and inhibit the proliferation of contaminating microorganisms. L-asparaginase can be used to treat cancer by depriving nutrients needed by the growth of cancer cells. Protease (like multi-enzyme tablets) can be used to treat dyspepsia, and it has an anti-inflammatory effect. In the world of bioprocessing, it is easy to fall into familiar assumptions: structure biologists rely on non-glycosylating E. coli systems, cost-conscious researchers favor low-cost yeast, biomanufacturers place their trust in the consistent quality of mammalian cells, such as CHO. However, while navigating in CHO upstream bioprocessing for decades now, I have learned that these clichés oversimplify a far more complex landscape. This recent paper from the Sarel Fleishman lab demonstrates one of the core technologies we will use in W-BioCat. In this paper, fungal enzymes that are extremely difficult to express heterologously were computationally designed to exhibit high stability and expression levels.
