#CustomerPublicationWeekly Immune infiltration within the tumor microenvironment (#TME) is important for immune checkpoint blockade (#ICB) therapy to function. Mild photothermal therapy (PTT) is often combined with immunotherapy to enhance efficacy, likely by increasing immune cell infiltration and stimulating T-cell activity. Notably, the PTT also accelerates the release of tumor cell-derived exosomes (TEX) and upregulates PD-L1 expression on both tumor cells and TEX. A multi-stage drug delivery system is designed that concurrently delivers photosensitizers (reduced graphene oxide nanosheets, NRGO), anti-PD-L1 antibodies, and exosome inhibitors (sulfisoxazole). This local combination therapy remodels the immunosuppressive TME through exosome inhibition and enhanced immune cell infiltration, while also boosting T-cell activity to trigger systemic antitumor immunity- making it a promising approach to further improve immunotherapy efficacy. Researchers from Luxembourg Centre for Systems Biomedicine presented a nigrostriatal pathway model based on midbrain-striatum #assembloids with inducible #aging. These assembloids can develop characteristics of the nigrostriatal connectivity, with catecholamine release from the midbrain to the striatum and synapse formation between midbrain and striatal neurons, making them excellent tools to reveal the contribution of aging as well as nigrostriatal connectivity to the onset and progression of Parkinson’s disease. #CustomerPublicationWeekly Erectile dysfunction (#ED) significantly impacts male #fertility and quality of life. A recent PNAS paper (https://lnkd.in/d84iZmgg) describes an innovative magnetic soft microrobot comprising an ultrasoft hydrogel microsphere embedded with a magnetic nanoparticle chain for mesenchymal stromal cells (#MSCs) delivery which shows significantly enhanced MSC retention and survival. In both rat and beagle ED models, MSC-Rob treatment accelerated the repair of corpus cavernosum tissue and restored erectile function. #Single-cell RNA sequencing (scRNA-seq) revealed that MSC-Rob treatment facilitates nerve and blood vessel regeneration in the corpus cavernosum by increasing the presence of regenerative macrophages. Singleron's GEXSCOPE Products were used in this study.

#CustomerPublicationWeekly Patients with gastric cancer peritoneal metastasis (#GCPM) have a poor #prognosis and respond poorly to conventional treatments. Recent development of immune checkpoint blockade (#ICB) has demonstrated favourable efficacy in the treatment of GCPM. A recent study published in the Gut magazine used #singlecellsequencing to analyse primary tumours, GCPMs and peripheral blood from patients with the aim to develop stratification strategy of of best responders. A stroma-myeloid crosstalk in GCPM is the major mediator of ICB resistance. Blocking the C3-C3AR1 axis disrupts the stroma-myeloid crosstalk and thereby significantly improves the benefits of ICB in in vivo models. The findings provide a comprehensive tumor microenvironment lanscape associated with ICB resistance in patients with GCPM and aid in the prioritisation of therapeutic candidates to potentiate immunotherapy.
