The Institute for Chemical Neuroscience (iCNS) has been awarded a 23.23 million euro #Gravitation grant. The funding will support research aimed at mapping changes in the brain, down to the smallest level, associated with psychiatric symptoms. Utrecht University professor Maarten Kole is one of the applicants of the project, which is a collaboration between universities, university medical centers, and the Netherlands Institute for Neuroscience. The overarching objective of the research is to improve the diagnosis and treatment of neuropsychiatric disorders such as depression, frontotemporal dementia, and anxiety disorders. Maarten Kole, who is also group leader at the Netherlands Institute for Neuroscience, has long been studying the role of glial cells: cells in our brains that, among other functions, provide support for nerve cells. Kole: “We believe that psychiatric symptoms are related to changes occurring in individual cells, and particularly between the glial cells and nerve cells. There is an increasing amount of brain material available from individuals with psychiatric disorders for investigating this issue. ☑️ *READ ASTRACT BELOW:* Keywords: Cellular biology; Molecular biology; Neuroscience; Techniques in neuroscience. Myelin defects cause a collection of myelin disorders in the brain. The lack of human models has limited us from better understanding pathological mechanisms of myelin diseases. While human induced pluripotent stem cell (hiPSC)-derived spheroids or organoids have been used to study brain development and disorders, it has been difficult to recapitulate mature myelination in these structures. Here, we have developed a method to generate three-dimensional (3D) myelin spheroids from hiPSCs in a robust and reproducible manner. Using this method, we generated myelin spheroids from patient iPSCs to model Canavan disease (CD), a demyelinating disorder. By using CD patient iPSC-derived myelin spheroids treated with N-acetyl-aspartate (NAA), we were able to recapitulate key pathological features of the disease and show that high-level NAA is sufficient to induce toxicity on myelin sheaths. Our study has established a 3D human cellular platform to model human myelin diseases for mechanistic studies and drug discovery. 🚀 Excited to announce our latest publication in eLife! 🚀 Our team has just published a study titled "Mapping Serotonergic Dynamics using Drug-Modulated Molecular Connectivity." This work represents a significant step forward in neuroscience, offering new insights into how drugs affect brain function at the molecular level. In our study, we utilized dynamic [11C]DASB PET scans to map serotonin transporters and examined the impact of MDMA (ecstasy) on these crucial mood regulators. Our findings reveal significant patterns of molecular connectivity changes post-MDMA. This innovative methodology not only enhances our understanding of brain function but also opens new avenues for exploring drug modulation in the brain. The diverse applications of BESA products in #neurophysiology are highlighted by the wide range of research from our customers. A quick review of recent publications has revealed exciting new trends. Here are a few highlights: ▶ Neural Oscillations and Statistics: Pastötter et al. used beamforming to localize increased post-movement beta synchronization in functional movement disorders. Brain Communications 6:5, 2024, fcae301 ▶ Connectivity and Visual Processing: Hu et al. studied brain responses to familiar vs. unfamiliar app icons, finding differences in the fusiform gyrus. NeuroReport 34:10, pp 521-525, 2023 ▶ Effects of Cannabis: Schantell et al. observed a marked suppression of gamma activity in specific brain regions in regular cannabis users. Human Brain Mapping 45:11, 2024 (e26787) ▶ Clinical Population Analysis: Busboom et al. explored motor performance in individuals with cerebral palsy, revealing that cognitive interference impacts beta and gamma oscillations, which in turn affects reaction times.
