🌐 AVC, the Association for Veterinary Consultants, was set up to provide a home for the community of independent and small-business regulatory specialists, covering the expertise-span from pre-clinical toxicology and field-trials to strategy and hands-on dossier management. Each year, the General Assembly is held in a different country and, in addition to providing the forum for statutory business, there is relaxed socialising and the opportunity to hear invited speakers, from amongst AVC’s members and from outside, addressing relevant topics. In October 2023 we gathered for our 28th GA in the warm and welcoming Mediterranean city of Nice, to mark also 10 years since taking up official residency in France. Nice also provided the backdrop for a discussion of Artificial Intelligence in Digital Pathology and Veterinary Toxicology. Research Update: I'm thrilled to share my latest publication on the pharmacokinetics of imipramine in dogs! This study sheds light on the unique pharmacokinetic behavior of this tricyclic antidepressant in a canine model, with some key insights that could inform future clinical applications in veterinary medicine 🐾🩺 Among the key highlights: - Rapid Absorption: Imipramine showed a remarkably fast absorption rate, considerably quicker than in other species like humans and horses. - Short Terminal Half-Life: The drug exhibited a very short terminal half-life of only 0.25 hours, highlighting the challenge of sustaining therapeutic plasma concentrations over time. This suggests the need for more frequent dosing or alternative formulations to achieve prolonged effects. Additionally, it's important to note that imipramine's half-life in humans is around 18 hours, making direct extrapolation of human dosing regimens to dogs potentially risky, as once-daily administration would likely be insufficient to maintain therapeutic levels in dogs. - Species-Specific Metabolism: We observed the formation of the active metabolite desipramine, but interestingly, it was not quantifiable in plasma. This points to species-specific differences in the metabolism of imipramine, which could influence its therapeutic efficacy in dogs compared to other species, given that desipramine is an active metabolite in humans (equivalent additive effect on efficacy). - Fasting vs. Fed Conditions: No significant differences in pharmacokinetics were observed between fasted and fed states, indicating that imipramine's bioavailability in dogs may not be influenced by food intake—a key consideration for its clinical use. Given imipramine's brief presence in therapeutic plasma concentrations, we recommend further research into sustained-release formulations and the use of therapeutic drug monitoring to optimize its application in veterinary medicine!! #VeterinaryMedicine #Pharmacokinetics #Research #AnimalHealth #DrugDevelopment #VeterinaryPharmacology #Canines

Here is a recent example of the risks of using unapproved or unregulated supplements in veterinary clinical trials. A paper published by Berman et al. (2024) in the Journal of Veterinary Pharmacology and Therapeutics shows this issue very clearly. The authors were planning to do a clinical trial to see if the botanical extract artemisinin could be used to treat Babesia gibsoni infections in dogs. Artemisinin is an antimalarial commonly used to treat the blood parasite Plasmodium falciparum in humans and has considerable therapeutic value in people.  Unfortunately for the authors they could only obtain nutraceutical forms of artemisinin and so before running their dog trial they conducted a small study to quantitate the levels of artemisinin in four common supplements using HPLC.  The results are surprising. One product had no artemisinin present and the other 3 had more than 150% of the stated label claim and all showed considerable variation between individual capsules. Any clinical trial with these products would have resulted in very questionable results.  Many supplements and nutraceuticals can suffer from this problem as they are unlikely to be manufactured to GMP standards. Any study using non-pharmaceutical grade ingredients is likely to have this same problem unless rigorous testing is performed.
